The Antibody Enigma

The Antibody Enigma

The Antibody Enigma is a somewhat personal view of the antibody diversity question from two investigators who have spent the past 18 years trying to penetrate the enigma. It is not and was not meant to be an all-embracing comprehensively referenced review of the subject of antibody diversity. Because of the subjective viewpoint, there are un doubtedly omissions of data that others consider to be seminal, and if we have offended anyone by omitting their own contribution we sin cerelyapologize. We have lived with "The Enigma" on and off for the past two years. It has been both hard work and good fun but, above all, it has been a learning experience. There were several difficult decisions to ma...

Antibody Engineering

Antibody Engineering comprises in vitro selection and modification of human antibodies including humanization of mouse antibodies for therapy, diagnosis, and research. This book comprises an overview about the generation of antibody diversity and essential techniques in antibody engineering: construction of immune, naive and synthetic libraries, all available in vitro display methods, humanization by chain shuffling, affinity maturation techniques, de novo synthesis of antibody genes, colony assays for library screening, construction of scFvs from hybridomas, and purification of monoclonal antibodies by exclusion chromatography. In addition, other topics that are discussed in this book are application and mechanism of single domain antibodies, structural diversity of antibodies, immune-mediated skin reactions induced by TNF-alpha recombinant antibodies, and bioinformatic approaches to select pathogen-derived peptide sequences for antibody targets.

The Variation and Adaptive Expression of Antibodies

During the past ten years, several theories have been proposed on the origin of the diversity of antibodies. George P. Smith presents a critical study of these theories in this detailed treatment of immunological problems from the point of view of molecular genetics. Mr. Smith uses a new and simplifying approach to this long-standing controversy. By a comprehensive computerized analysis of antibody amino acid sequences (particularly the myeloma proteins), the author traces their evolution and matches his results against the expectations of the various theories of diversity. He discusses at length the other types of evidence as well. Mr. Smith also deals with the clonal specialization of cells to produce a single antibody, and the relationship of this specialization to the somatic joining of antibody half-genes, which is one of the immune system's most important peculiarities. Introductory material is provided to make this work understandable to molecular geneticists not versed in immunology and to immunologists not versed in molecular genetics. This is a timely book offering a succinct and coherent summary of the various lines of evidence in a confused and controversial field.

The Generation of Diversity

This book is an intellectual history of the major theoretical problem in immunology and its resolution in the post-World War II period. In recent years immunology has been one of the most exciting--and successful--fields of biomedical research; this book provides essential background for understanding the conceptual conflicts occurring in the field.

Somatic Diversification of Immune Responses

Discovery of the mechanism for V(D)J hypermutation remains a basic goal of immunology despite the best efforts of many labo ratories. The existence of catalyzed, site-specific mutation and its exploitation for the somatic evolution of lymphocytes are re markable adaptations, yet since the discovery of hypermutation in 1970 (see cover), much hard work has generated little. Indeed, our knowledge of what is probably absolutely required for the mutator's action can be succinctly expressed: /g gene enhancers. Table 1 of Winter et a!.'s chapter puts into a historical perspec tive how our notions of the mutator have changed over the years. Despite these modest gains, most of us feel that this is the best of times. Our work has not only shown us what the mutator is not, it has also, like an artist's preliminary sketch, defined the questions and experiments we must face without diminishing the potential for new biology. In short, it is great fun to toil against a significant and enigmatic problem.

This contemporary book covers significant new knowledge that has emerged during the last two decades and, thus, provides novel antibody phylogenetic perspectives relevant to development of new antibody-based therapeutics and vaccines. It fills a much-needed niche in the area of immunoglobulin genetics across species from a comparative perspective. New insights and perspectives from immunoglobulin genetics from species such as sea lamprey, cattle, marsupial, bat, rat, rabbit, and swine—other species than the traditional subjects of mice and humans—are relevant to antibody design and engineering. These new perspectives find direct application in the cutting-edge areas of antibody design, e...

A practical guide to the production of antibodies. Describes the biosynthesis of antibody molecules by B lymphocytes and their progeny, the use of genetic markers in the elucidation of mechanisms of antibody synthesis, and of the generation of antibody diversity. Also deals with anomalies in antibody production which result in immunoglobulinopathies, involving qualitative or quantitative abnormalities, or both, in the immunoglobulins.

Fc Mediated Activity of Antibodies

Fc Mediated Activity of Antibodies

This volume explores several aspects of how antibodies mediate their activity in vivo, ranging from cancer immunotherapy to autoimmunity, infection, and vaccination. Divided into seven chapters, it provides in-depth insights into how antibodies and especially the antibody fragment crystallizable (Fc) domain modulate immune responses and antibody activity. The book begins by discussing evolutionary aspects of how the family of Fc receptors that are the key molecules for antibody activity evolved. In turn, it addresses the molecular and cellular pathways underlying IgG activity in cancer immunotherapy, and focuses on how IgG glycosylation regulates IgG and IgE activity in autoimmunity, allergy and infection. In closing, it presents strategies for developing novel antibody-based vaccination approaches. The book is intended for a very broad readership, including graduate students, postdocs and principal investigators with a basic grasp of immunology.
