This translational, clinically oriented book describes in detail novel approaches to cancer immunotherapy, current strategies to target tumor immunosuppression, and prognostic biomarkers for personalized cancer treatments. Since the first, very successful edition of the book was published in 2015, the original chapters have been significantly updated and entirely new chapters are included on, for example, cancer immunoprevention, aptamer-mediated cancer gene therapy, haploidentical bone marrow transplantation for pediatric malignancies, and nanoimmunotherapy. This book provides a comprehensive overview of the importance of molecular imaging in multiple myeloma, with detailed explanation of its clinical impact. Other important features are the definition of criteria that will aid PET/CT interpretation; identification and explanation of the most frequent pitfalls; a brief overview of the advantages and limitations of DWI MR imaging, still an experimental technique in multiple myeloma; and examination of the possible role of emerging PET tracers. When appropriate, clinical cases are used to illustrate key teaching points. All physicians involved in oncological imaging should regularly reassess and update their routine practice in the evaluation of multiple myeloma patients. This is especially true now, given the recent clarification by the International Myeloma Working Group (IMWG) of the criteria for bone damage requiring therapy and the emerging data supporting the role of the newer functional imaging techniques in predicting outcome and/or evaluating response to therapy. In medicine, endocrinology is perhaps the domain that encompasses the most molecular imaging using radioactive agents, also called radiopharmaceuticals. This is due to the diversity of molecular targets, including hormone synthesis pathways expressed uniquely by endocrine tumors and the growing knowledge in their pathophysiology and genetics. Those radiopharmaceuticals not only serve as diagnostic agents, but can also serve as a platform for characterizing endocrine disorders and treat patients and be used to monitor intracellular events that are induced by various therapies. Finally, they may be able to predict the aggressiveness and metastatic potential of endocrine tumors. This book provides comprehensive and up-to-date insights into the diagnostic, therapeutic, and future approaches of nuclear medicine to endocrine tumors in a new spirit of precision medicine. Improved understanding of the cellular and molecular makeup of tumors in the last 30 years has unraveled a previously unexpected level of heterogeneity among tumor cells as well as within the tumor microenvironment. The concept of tumor heterogeneity underlines the realization that different tumors can display significant differences in their genomic content as well as in their overall behavior. Our capacity to better understand the heterogeneous make up of tumors has very important consequences on our ability to design efficient therapeutic strategies to improve patient survival. This book highlights several aspects of tumor heterogeneity in the context of metastatic development and summarize some of the challenges posed by heterogeneity for tumor diagnostics and therapeutic management of tumors.
The aim of this Research Topic was to assemble a series of articles describing basic, preclinical and clinical research studies on radiopharmaceuticals and nuclear medicine. The articles were written by attendees of the third Nuclear Technologies for Health Symposium (NTHS, 10th-11th March 2015, Nantes, Frances) under the auspices of the IRON LabEx (Innovative Radiopharmaceuticals for Oncology and Neurology Laboratory of Excellence). This French network, gathering approximately 160 scientists from 12 academic research teams (Funded by “investissements d’Avenir”), fosters transdisciplinary projects between teams with expertise in chemistry, radiochemistry, radiopharmacy, formulation, biology, nuclear medicine and medical physics. The 12 articles within this resulting eBook present a series of comprehensive reviews and original research papers on multimodality imaging and targeted radionuclide therapy; illustrating the different facets of studies currently conducted in these domains.
