The development of new CNS drugs is notoriously difficult. Drugs must reach CNS target sites for action and these sites are protected by a number of barriers, the most important being the blood –brain barrier (BBB). Many factors are therefore critical to consider for CNS drug delivery, e.g. active/passive transport across the BBB, intra-brain distribution, and central/systemic pharmacokinetics, to name a few. Neurological disease and trauma conditions add further complexity because CNS barriers, drug distribution and pharmacokinetics are dynamic and often changed by disease/trauma. Knowledge of all these factors and their interplay in different conditions is of utmost importance for proper...

Volume 1 of this series focused mainly on small molecules which are intrinsically electroactive. Volume 2 continues to provide current approaches to real time measurements of essential species in the central nervous system. It describes microdialysis, a sampling technique, which facilitates the removal of small volumes of solution containing the analyte(s) of interest which can be subsequently separated by chromatography and analyzed by a range of techniques. These include electrochemistry, UV-Vis spectroscopy, fluorescence and mass spectroscopy. This approach affords the opportunity to measure a range of targets in real-time thus providing access to pharmacokinetic/pharmacodynamics measurem...
A vast number of diagnostic and therapeutic decisions are based on measuring blood concentrations of molecules, yet most biochemical and pharmacological events actually take place in the tissues. Microdialysis is a key semi-invasive sampling technique to measure in vivo drug penetration to the target site in humans, the method being feasible in virtually every organ. Authored by international experts in this cutting edge field, this book will provide a comprehensive overview of microdialysis and its application for measuring drug distribution in drug development.
Medicinal chemistry is both science and art. The science of medicinal chemistry offers mankind one of its best hopes for improving the quality of life. The art of medicinal chemistry continues to challenge its practitioners with the need for both intuition and experience to discover new drugs. Hence sharing the experience of drug research is uniquely beneficial to the field of medicinal chemistry. Drug research requires interdisciplinary team-work at the interface between chemistry, biology and medicine. Therefore, the topic-related series Topics in Medicinal Chemistry covers all relevant aspects of drug research, e.g. pathobiochemistry of diseases, identification and validation of (emerging...
This publication contains the speeches and conclusions of a seminar held in October 1999, which looked at the role of the pharmacist as a co-guarantor of health security. The first theme was health challenges of the 21st Century, which included the safety of new therapies, the pharmacists role in risk management and the problems of the counterfeiting of drugs. The second theme concerned the challenges of the new technologies both the dangers of selling drugs over the internet and the opportunities of increased networking professional information. The final theme looked at the risks of the new technologies and the ways that the pharmacist could add value. The conclusions of the seminar will serve as the framework for a Resolution of the Council of Europe's Committee of Ministers.
This book presents a comprehensive collection of current knowledge and leading research about the blood-brain barrier. The chapters are organized in four main parts providing basic information and novel insights about the physiology of the blood-brain barrier, the challenges related to finding and developing drugs crossing the blood-brain barrier, experimental methods to study the blood-brain barrier and the role of the blood-brain barrier in disease mechanisms and its consequences for drug development. In the first part the readers will discover the structure, function and developmental aspects of the blood-brain barrier and gain novel insights into the complexity and functionality of the n...
Microdialysis is currently one of the most important in vivo sampling methods in physiology and pharmacology. It is used to determine the chemical components of the fluid in the extracellular space of tissues. The technique is now well established in neuroscience research and is used excessively in behavioral neuroscience to determine the concentrations and identities of molecules in brain tissues, and their change due to behavior, hormonal and transmitter changes in the nervous system. The book provides a detailed comprehensive overview of the technology and its applications, including application in pathology, drug development, and the application in the clinic. The authors are all well kn...
