The past two decades have seen steady progress in our understanding of the pathogenesis of atherosclerosis. The role of low density lipoprotein (LOL) increase and of LOL receptor deficiency or malfunctions in familial hypercholesterolemia has been largely enlightened by the works of Brown and Goldstein. These authors postulated also that modification of LOL to a form recognized by the scavenger or acetyl-LOL receptor may be required for lipid loading of macrophage-derived foam cells in the lesions. A growing body of evidence suggests that oxidative modification of LOL could enhance its atherogenicity by its implication as a factor in the generation of foam cells. Low Density Lipoproteins

Low density lipoproteins (LDL) are pathophysiologically important be cause of their central role in the disease atherosclerosis and because atherosclerosis is the leading cause of death in developed countries. Many researchers believe that a more detailed knowledge of the struc ture, function, and metabolism of LDL may eventually lead to a means to control atherosclerosis. For this reason a fairly large research effort has gone into the investigation of LDL over the past few years. The purpose of this book is to collect and summarize in one place most of the pub lished information on LDL through 1975. To this end more than 1500 references are cited in the papers that make up this volume. The A, B, C apolipoprotein classification system was adopted for use throughout this work. In addition to the A, B, C, and "D" families of apolipoproteins, apoE is used to designate the "arginine-rich" apolipo protein. This classification system is used because it is far less cumber some than other proposed classification schemes for apolipoproteins.

Pharmacological Control of Lipid Metabolism

This Symposium was the fourth in a series which began in Milan, Italy, in 1960. Each meeting has introduced or developed some new concepts in the areas of lipid metabolism and drugs. The meetings have served as a springboard for new ideas which have, between meetings, become accepted and exploited. This meeting has been no exception. Principal among the many new concepts discussed were lipoprotein synthesis and metabolism, apoprotein structure and function, whole body metabolism of cholesterol, and aspects of myocardial and aortic metabolism. The Symposium also included a summary of current thought on management of hyperlipemias and atherosclerosis. Data on more than 30 drugs were introduced and discussed. We have every expectation that the next Symposium will include material which is now only in the formative stage.
