Medicinal chemistry is both science and art. The science of medicinal chemistry offers mankind one of its best hopes for improving the quality of life. The art of medicinal chemistry continues to challenge its practitioners with the need for both intuition and experience to discover new drugs. Hence sharing the experience of drug research is uniquely beneficial to the field of medicinal chemistry. Drug research requires interdisciplinary team-work at the interface between chemistry, biology and medicine. Therefore, the topic-related series Topics in Medicinal Chemistry covers all relevant aspects of drug research, e.g. pathobiochemistry of diseases, identification and validation of (emerging...

Resolving the Antibiotic Paradox

Antibiotic resistance, once a term appreciated only by microbiologists, has become a common topic in the popular press. Stuart Levy, one of the contributors to this collection, bears some of the responsibility for increasing public awareness with the publication of his book "The Antibiotic Paradox" in 1992: Misuse of antibiotics resulting in increased bacterial resistance had previously been recognized in the infectious disease community. However, Dr. Levy's eloquent public warning about the shrinking efficacy of our antibi otic armamentarium served to alert the lay person to the potential consequences of this de mise in useful therapy. Because of the proliferation of diverse antibiotic clas...

The enormous genetic flexibility of bacteria jeopardizes the usefulness of currently available antibiotics, and requires new approaches to antibiotic discovery and development. Antimicrobial resistance can be acquired in a short time frame, both by genetic mutation and by direct transfer of resistance genes across genus and species boundaries. Unde

Technological Challenges in Antibiotic Discovery and Development

Technological Challenges in Antibiotic Discovery and Development is the summary of a workshop convened by the Chemical Sciences Roundtable in September 2013 to explore the current state of antibiotic discovery and examine the technology available to facilitate development. Through formal presentations and panel discussions, participants from academia, industry, federal research agencies discussed the technical challenges present and the incentives and disincentives industry faces in antibiotic development, and identified novel approaches to antibiotic discovery. Antibiotic resistance is a serious and growing problem in modern medicine and it is emerging as a pre-eminent public health threat....

Wound Healing

Outstanding scientific advances over the last decades unceasingly reveal real complexity of wound-healing process, astonishing in its staged progression, as life is unfolding itself. This natural course of tissue repair seems to bear thousands of overlapping molecular and macroscopic processes that nowadays only start to unfold to our knowledge. The present volume collecting recent scientific references proposes to readers a two-folded audacious goal. First, an updated design of intimate cellular mechanisms is entailed in tissue regeneration that emanates from the first section of the book. Next, a multidisciplinary therapeutic perspective that focuses on macroscopic healing throughout the s...

Antimicrobials

Reports on the emergence and prevalence of resistant bacterial infections in hospitals and communities raise concerns that we may soon no longer be able to rely on antibiotics as a way to control infectious diseases. Effective medical care would require the constant introduction of novel antibiotics to keep up in the “arms race” with resistant pathogens. This book closely examines the latest developments in the field of antibacterial research and development. It starts with an overview of the growing prevalence of resistant Gram-positive and Gram-negative pathogens, including their various resistance mechanisms, prevalence, risk factors and therapeutic options. The focus then shifts to a comprehensive description of all major chemical classes with antibacterial properties, their chemistry, mode of action, and the generation of analogs; information that provides the basis for the design of improved molecules to defeat microbial infections and combat the emerging resistances. In closing, recently developed compounds already in clinical use, those in preclinical or first clinical studies, and a number of promising targets to be exploited in the discovery stage are discussed.

Activating Agents and Protecting Groups
