Human Tumor-Derived p53 Mutants: A Growing Family of Oncoproteins

TP53 gene mutations are present in more than half of all human cancers. The resulting proteins are mostly full-length with a single amino acid change and are abundantly expressed in cancer cells. Some of the mutant p53 proteins gain oncogenic functions (GOF) through which it actively contribute to the aberrant cell proliferation, increased resistance to apoptotic stimuli and ability to metastasize. Gain of function mutant p53 proteins can transcriptionally regulate the expression of a large plethora of target genes. This mainly occurs through the formation of oncogenic transcriptional competent complexes that include mutant p53 protein, known transcription factors, posttranslational modifier...

Our understanding of human cancer in the past 40 years has been driven by linking innovative concepts and cutting edge technologies to key problems identified by clinical research. Some of the successes in cancer genetics identified from clinical work have been the identification of specific gene deletions in human chromosomes, the use of PCR-based cloning methodologies to identify and clone human cancer genes, the validation of the human cancer genes using transgenetic technologies in the mouse, and the ability to sequence whole genomes that has recently allowed a collation of all somatic and germline mutations in a human genome. In the same generation, entirely different disciplines involv...

This book provides the readers with an overview of research on p53, which has been shown to play a role in numerous crucial biological pathways in normal and cancer cells. Leading scientist in the field, who have all made direct contributions to the understanding of the molecular events underpinning p53 function, have been invited to contribute the various chapters, which discuss the current knowledge of the signaling cascades that are activated by mutations in p53 and overexpression of MDM2, frequently found in human cancer and are major causes of oncogenesis. This book features chapters on the molecular basis of oncogenesis induced by gain of function mutation of p53, signaling pathways induced by MDM2 overexpression, control of mutant or wild-type p53 function by MDM2 and MDMX, p53 mutation in hereditary cancer and structural aspects that activate mutant p53 which can be targeted by drug therapy. This book should be useful for scientists at all levels.

Ecological and Evolutionary Genetics of Drosophila

Ecological and evolutionary genetics span many disciplines and virtually all levels of biological investigation, from the genetic information itself to the principles governing the complex organization of living things. The ideas and informa tion generated by ecological and evolutionary genetics provide the substance for strong inferences on the origins, changes and patterns of structural and functional organization in bio logical communi ties. It is the coordination of these ideas and thoughts that will provide the answers to many fundamental questions in biology. There is no doubt that Drosophilids provide strong model systems amenable to experimental manipulation and useful for testing pe...

Cell Death

This book on cell death contains 29 self-contained, peer-reviewed articles written by leading scientists in each field. It features overview articles aimed at undergraduates and non-specialists, which present basic information and provide entry into the following advanced articles. These advanced articles are written for postgraduate students and research workers, containing detailed information and key references allowing the reader to investigate a specific area in more depth. The book is an essential resource for educational purposes as well as a reference work for experienced researchers in the field. The articles will also be available electronically as part of the acclaimed Encyclopedi...

AACR 2016: Abstracts 2697-5293

The AACR Annual Meeting is a must-attend event for cancer researchers and the broader cancer community. This year's theme, "Delivering Cures Through Cancer Science," reinforces the inextricable link between research and advances in patient care. The theme will be evident throughout the meeting as the latest, most exciting discoveries are presented in every area of cancer research. There will be a number of presentations that include exciting new data from cutting-edge clinical trials as well as companion presentations that spotlight the science behind the trials and implications for delivering improved care to patients. This book contains abstracts 2697-5293 presented on April 19-20, 2016, at the AACR Annual Meeting.

p53 Protocols

Since the discovery of p53 as a tumor suppressor, numerous methods have evolved to reveal the unique structural features and biochemical functions of this protein. Several unique properties of p53 posed a challenge to understa- ing its normal function in the initial phase of its research. The low levels of p53 in normal cells, its stabilization under situations of genotoxic stress, induction of growth arrest, and apoptosis with stabilization of the protein, obstructed the visibility of its normal, unmutated function. The property of p53 that can sense a promoter and transactivate or inhibit is still not well understood. It is still not known whether it is the absence of the protein that caus...
