IL-1 Inhibition
This special article collection of Frontiers in Pharmacology includes reviews and original articles on different aspects of IL-1 inhibition. Since the time IL-1 and its natural antagonist IL-1Ra have been discovered, specific IL-1 targeted therapies have been developed to cure an increasing number of diseases. IL-1ra in the Treatment of Rheumatoid Arthritis: pocketbook
In recent years researchers have gained a greater understanding of the immunological and inflammatory mechanisms underlying rheumatoid arthritis. Much of their efforts have centred on the role of the cytokine network in the pathophysiology of the disease. On the basis of these discoveries pharmaceutical companies have developed several potential biologic agents aimed at addressing cytokine imbalance and its subsequent effects on cartilage damage and joint destruction in rheumatoid arthritis. This pocketbook will provide an overview of the role of cytokines in the disease process, with a particular focus on one cytokine, interleukin-1 (IL-1) and on the therapeutic efficacy of its inhibition.

T Cells in Arthritis
Rheumatoid arthritis (RA) is the most common and most severe form of inflammatory arthritis. The pathogenesis of RA has been the subject of intense research for several decades. The prevailing hypotheses have changed over the years, and have attempted to incorporate the most recent data. Although T cells represent an important component of the cells which infiltrate the joint synovium, their contribution at a late stage of the disease remains a matter of debate. The goal of this book is to outline the major arguments and data suggesting that T cells may, or may not, be central players in the pathogenesis of chronic RA. While each of the editors and authors has his/her own bias (as will be clear by reading the respective chapters), our hope is that the readers will enjoy a complete and balanced view of the critical questions and experiments. This is not just an intellectual exercise since the direction of future therapeutic interventions depends heavily on how one interprets the pathogenesis of RA and the contribution of T cells.
