The voluminous literature on prostaglandins includes some excellent reviews. For some time however, there has been a need for the collection in one volume of information from individual experts actively engaged in different aspects of prostaglandin research. This book is an attempt to fulfil that need. This is the second in a series of three books on advances in prostaglandin research. In recent years there has been an unparalleled interest in these compounds and as a result a vast amount of research data has accumulated since the publication of my earlier book in 1972. At that time it was possible to present a fairly comprehensive review of the various aspects of prosta glandin research in one volume. In June, 1981, we conducted a two day international symposium in Rome devoted to original scientific presentations on arachidonic acid metabolism in the kidney. Scientists from 20 countries were represented either in the general scientific program or in the poster presentation. This book does not represent the proceedings of this international gathering, but rather the edited manuscripts specifically prepared as sum maries of the scientific presentations. This book was written at the invitation of Dr. H. GOTZE of Springer-Verlag for the series "Monographs in Endocrinology". It is not a comprehensive account of the prostaglandins but has been writ ten with a deliberate emphasis upon those aspects of the field in which I am particularly interested and to which, in some cases, I have made a contribution. I am grateful to Miss E. PFISTERER and her colleagues of Springer Verlag for their excellent work. I should also like to thank my wife without whose patience, encouragement and help this book would never have been completed. The introduction of prostaglandins into clinical practice has been one of the most important advances in obstetrics and gynaecology in recent years. During the last decade obstetricians have become familiar with these drugs for inducing labour and for terminating second-trimester pregnancy. Within the last year their use has been extended, in association with antiprogestin, to first-trimester termi nation. Although the effectiveness of prostaglandins in these pharma cological roles is clear, their full potential has still to be explored, and much remains to be learned about their physiology. Prostaglandins playa central role in the initiation of labour. Further clarification of this role could lead to improvements in preventing or treating preterm labour, which still remains the most important cause of perinatal mortality in this country. Prostaglandins are also inti mately involved in the mechanism of menstruation. More detailed understanding of this role should lead to more effective treatments for menorrhagia and dysmenorrhoea. These substances also playa part in the mechanism of implantation, and further research in this area may lead to more effective therapy for infertility.
Prostaglandin Synthesis reviews the developments in the preparation and isolation of prostaglandins by synthesis, with emphasis on the more promising and significant approaches. It includes a nomenclature of prostaglandins and considers prostaglandin analogs as well as their structure-activity relationships, prostaglandin interconversions, isomeric prostaglandins, and deoxyprostaglandins. Organized into 21 chapters, this book begins with an overview of prostaglandin biosynthesis and the potential clinical utility of prostaglandins. Since prostaglandins were first proposed as mediators of in flammation some eight years ago, there have been a variety of arachidonic acid metabolites discovered including the thromboxa nes, prostacyclin, prostaglandin endoperoxides and lipoxygenase products such as SRS-A.
