This book focuses on an “outside the box” notion by utilizing the powerful applications of next-generation sequencing (NGS) technologies in the interface of chemistry and biology. In personalized medicine, developing small molecules targeting a specific genomic sequence is an attractive goal. N-methylpyrrole (P)–N-methylimidazole (I) polyamides (PIPs) are a class of small molecule that can bind to the DNA minor groove. First, a cost-effective NGS (ion torrent platform)-based Bind-n-Seq was developed to identify the binding specificity of PIP conjugates in a randomized DNA library. Their biological influences rely primarily on selective DNA binding affinity, so it is important to analyz...

Synthesis and Biological Evaluation of Pyrrole–Imidazole Polyamide Probes for Visualization of Telomeres

This book presents a novel method of synthesizing sequence-specific DNA binding pyrrole–imidazole (Py–Im) polyamides and demonstrates polyamides targeting longer base pairs than those previously reported and their cellular application. It discusses the development of the facile synthetic method of tandem hairpin Py–Im polyamides and the method of visualizing telomeres under mild conditions without harsh denaturation, as well as research resulting in the synthesis not only of previously reported tandem dimer polyamides but also longer tandem trimer and tetramer polyamides. Using fluorescently labeled tandem hairpin Py–Im polyamides targeting a human telomere repetitive sequence, the author successfully stained telomeres in fixed cells without denaturation. These fluorescently labeled tandem tetramer polyamides are able to visualize human telomeres with lower background signals than previous polyamide probes, suggesting that they have higher specificity for telomeres. As such, this book provides insights into recent advance in Py–Im polyamides, particularly the extension of the target sites c

Development of Selective DNA-Interacting Ligands

This book addresses the development of both DNA-sequence-selective and DNA-form-selective ligands, with the aim of creating potential molecular probes and therapeutic agents for non-canonical DNA structure-caused human diseases. Over the past two decades, the structural diversity of DNA forms has been proven to have profound implications in various biological, neurological, and pharmacological events. In response, researchers have since made tremendous efforts to obtain highly active drugs interacting with disease-related non-canonical DNA structures. These drugs, however, have not yet been approved for clinical use. One obstacle impeding their clinical application has to do with selectivity...

Lions 324B1 District Directory (2013-14)

Print Edition of Lions District 324B1 Directory for 2013-14 was released by District Governor MJF Lion S Shanmugam in August 2013, during his regime. Considering the developments in Mobile Technology, Digital Directories were introduced for Lionism in November 2015, by Lion Dr Er J Shivakumaar. To create Archives of Lions Directories Digitally and make available in every Lion's Mobile Phones, This Edition is Digitised in September 2016. This is replica of the Print Edition and enables availability of information on previous years and will serve as a reference source.
