Acute and Hereditary Coagulation Disorders
Coagulation disorders can have a variety of causes and may lead to either an increased risk of excessive bleeding or an increased risk of thromboembolic events. Both types of coagulation disorders can be life-threatening and require immediate therapy. Major bleeding is often triggered by an acute cause, such as severe trauma or peripartum hemorrhage (PPH) . The high rate of pre-existing intake of anticoagulants, such as DOACS or Vitamin-K antagonists, can also induce or even aggravate bleeding events, for example in cerebral hemorrhage or traumatic brain injury (TBI). Hereditary causes, such as the congenital Factor XIII deficiency or hemophilia, can also trigger a clinically relevant hemorrhage however, hereditary coagulation disorders are more often associated with thromboembolic events.
Analysis and improvement of the in vitro transfection efficiency of plasmid-DNA encoding for equine IL-12 used for melanoma therapy in horses
In horses studies targeting equine melanoma therapy using DNA encoding for cytokines as interleukin 12 (IL-12) have shown some promising results. Judith Krawinkel zeigt anhand von Goldnanostrukturen, dass deren Interaktion mit Laserstrahlung das Potenzial hat, schonend und effizient Wirkstoffe in Zellen einzubringen, diese gezielt zu eliminieren oder genetisch zu manipulieren. So eignen sich membranadhärente sphärische Goldnanopartikel zum Einbringen von Molekülen in Zellen. Vor allem bei Primärzellen empfiehlt die Autorin die Zugabe von Radikalfängern. Zudem nutzt sie peptidkonjugierte Goldnanopartikelagglomerate, die die Aufnahme extrazellulärer Moleküle über Endozytose unterstützen. Laserbestrahlung bewirkt effizientes intrazelluläres Freisetzen der Moleküle und der Agglomerate, die dabei in biokompatiblere Produkte zerteilt werden. Durch Bestrahlen goldbeschichteter Oberflächen mit Lasern entstehen Nanopartikel und Kavitationsblasen, die eukaryotische Zellen irreversibel schädigen und Biofilme manipulieren können.
