Optical microendoscopy is an emerging modality for imaging in live subjects. Using gradient refractive index (GRIN) microlenses, microendoscopy enables subcellular-resolution imaging in deep tissues that are inaccessible by traditional imaging techniques. We present a platform of methods and technologies that build upon GRIN microendoscopy: 1) miniaturized microscopes for imaging in awake, behaving animals, 2) methods for imaging contractile dynamics in the muscles of animal and human subjects, 3) chronic brain preparations that allow for longitudinal examinations of subcellular neuronal features and disease progression, and 4) novel microendoscope probes whose imaging capabilities approach that of standard water-immersion microscope objectives. When combined with the broad sets of available fluorescent reporters, and minimally invasive surgical preparations, the work described in this dissertation enables sophisticated experimental designs for probing how cellular char- acteristics may underlie or explain behavior, in models of both healthy and diseased states.
Optical microresonators have generated much progress in many aspects of optical science, such as in optical buffering, wavelength division-multiplexed filters for optical networks, optical switching, and in the enhancement of nonlinear effects. All these applications are made possible by the confinement of light within a small modal volume in the microresonator. The ability to actively tune the resonance within a microresonator has resulted in systems that can dynamically manipulate the flow of light. This thesis discusses some of the novel capabilities of these dynamic systems. Chapters 3 - 4 discusses the use of a dynamic delay line to suppress dispersion, and to overcome the gain-bandwidth product constraint of slow-light structures. This is followed by Chapter 5 which introduces a light-stopping scheme based on loss modulation. Finally, Chapter 6 shows how coherent control can be used in a microresonator system to enhance optical switching.

Micromachined Mirrors provides an overview of the performance enhancements that will be realized by miniaturizing scanning mirrors like those used for laser printers and barcode scanners, and the newly enabled applications, including raster-scanning projection video displays and compact, high-speed fiber-optic components. There are a wide variety of methods used to fabricate micromachined mirrors - each with its advantages and disadvantages. There are, however, performance criteria common to mirrors made from any of these fabrication processes. For example, optical resolution is related to the mirror aperture, the mirror flatness, and the scan angle. Micromachined Mirrors provides a framework for the design of micromirrors, and derives equations showing the fundamental limits for micromirror performance. These limits provide the micromirror designer tools with which to determine the acceptable mirror geometries, and to quickly and easily determine the range of possible mirror optical resolution and scan speed.
