The Nobel Prize in Medicine 1968 for interpretation of the genetic code and its function in protein synthesis and in Chemistry 2009 for studies of the structure and function of the ribosome highlighted the ground-breaking experiment performed on May 15, 1961 by Nirenberg and Matthaei and their principal breakthrough on the creation of "cell-free protein synthesis (CFPS) system". Since then the continuous technical advances have revitalized CFPS system as a simple and powerful technology platform for industrial and high-throughput protein production. CFPS yields exceed grams protein per liter reaction volume and offer several advantages including the ability to easily manipulate the reaction components and conditions to favor protein synthesis, decreased sensitivity to product toxicity, batch reactions last for multiple hours, costs have been reduced orders of magnitude, and suitability for miniaturization and high-throughput applications. With these advantages, there is continuous increasing interest in CFPS system among biotechnologists, molecular biologists and medical or pharmacologists.
Enzyme inactivation in fruits and vegetables is of utmost importance regarding food quality during storage. This new volume explores important emerging technologies for the inactivation of enzymes in the design and preservation of food. The current advances in sequencing, data mining, DNA synthesis, cloning, in silico modeling, and genome editing have opened a new field of research known as Synthetic Genomics. This has opened the possibility to further improve our understanding of genome fundamentals by considering the effect of the whole biological system on biological function. This book is divided into five sections. The first section deals with the methodology and bioresource generation, techniques related to genetic engineering, and gene transfer to the nuclear genome and chloroplast genome. The new techniques of genome profiling and gene silencing are also presented. The second section of the book covers the classical aspect of plant biotechnology viz. tissue culture and micropropagation. Use of genetic engineering via Agrobacterium and direct transfer of DNA through particle bombardment to develop transformed plants in Artemisia, castor and orchids, and production of recombinant proteins in plant cells have been dealt with in the third section. The fourth section addresses the abiotic and biotic stress tolerance in plants.
