DNA damage is a major threat to genomic integrity and cell survival. It can arise both spontaneously and in response to exogenous agents. DNA damage can attack most parts of the DNA structure, ranging from minor and major chemical modifications, to single-strand breaks (SSBs) and gaps, to full double-strand breaks (DSBs). If DNA injuries are mis-repaired or unrepaired, they may ultimately result in mutations or wider-scale genome aberrations that threaten cell homeostasis. Consequently, the cells elicit an elaborate signalling network, known as DNA damage response (DDR), to detect and repair these cytotoxic lesions. This Research Topic was aimed at comprehensive investigations of basic and novel mechanisms that underlie the DNA damage response in eukaryotes.

Telomeres are specialized DNA–protein structures that protect the ends of chromosomes to maintain their integrity. They are chromosome sentinels. This book, written by multiple authors, presents current knowledge and the most recent discoveries relating to telomeres. The first chapters describe telomere structures (DNA and proteins), organization, dynamics and replication in a range of organisms including ciliates, yeast and mammals. The following chapters cover mechanisms that maintain telomere length involving telomerase and Alternative Lengthening Telomeres (ALT) mechanisms. Interestingly, telomeres are transcribed into repetitive RNA called TERRA (Telomeric repeat-containing RNA), the state of the art regarding mammalian telomere transcripts is presented here. Two chapters are dedicated to the description of how telomere biology impacts the development of cancer and how their disfunctions induce telomeropathies (telomere biology disorders). Finally, the book ends with a chapter presenting telomere and telomerase targeting agents as potential anticancer agents, a promising therapeutic approach.

This volume presents the most current reviews on how cancer stem cells (CSCs) hypothesis dictates that the continued proliferation of a tumor is dependent on a sub-population of self-renewing and asymmetrically dividing neoplastic stem cells that supply a largely differentiated tumor. This volume provides a comprehensive overview of the characteristics of CSCs, their role in central nervous system (CNS) tumors, and the recent CSC-specific treatment modalities being used. The emerging focus on CSCs in brain tumors represents a paradigm shift in our understanding of the pathogenesis of these neoplasms. Importantly, the realization that a distinct sub-population of cells contributes disproportionately to the growth and sustenance of central nervous system tumors has important implications for the treatment of such tumors. To treat CNS tumors, there is now a growing need to treat CSCs to achieve adequate tumor control.
