Challenges Associated with Identifying Preclinical Animal Models for the Development of Immune-based Therapies
There hasn’t been a better time to be a drug developer for immune-based therapies than the past couple of decades. We have seen an explosion in immune-based therapies for cancer, autoimmune and infectious diseases, metabolic diseases and diseases and disorders of the nervous system. The modalities of these immune-based therapies span small molecules, biologics, and gene and cell therapeutic approaches. Significant advances have been made in optimizing drug design for its specificity for the target, characterizing the mode of action in in vitro assays, and ensuring safety and manufacturability. However, an area of challenge that remains is identifying animal models for evaluating efficacy and pharmacokinetics/pharmacodynamics relationship that are predictive of drug effects in humans. Discussion on this topic is warranted as examples of failures of translation from animal models to humans provide us an opportunity to learn more about human biology.
Case Reports in Thoracic Oncology: 2022
This Research Topic aims to collect all the Case Reports submitted to the Thoracic Oncology section. All the Case Reports submitted to this collection will be personally assessed by a senior Associate Editor before the beginning of the peer-review process. Please make sure your article adheres to the following guidelines before submitting it. Case Reports highlight unique cases of patients that present with an unexpected diagnosis, treatment outcome, or clinical course. Only Case Reports that are original and significantly advance the field will be considered: 1) RARE case with TYPICAL features. 2) FREQUENT case with ATYPICAL features. 3) Cases with a convincing response to new treatments, i.e. single case of off-label use.
Integrative Pharmacology-based Research on Traditional Medicine: Methodologies, Medical and Pharmacological Applications
ntegrative Pharmacology can be used to determine the multi-pharmacological effects of traditional medicines such as traditional Chinese medicine (TCM), Kampo, Sa-sang, Ayurveda, etc.). Through qualitative and quantitative pharmacokinetic-pharmacodynamic (PK-PD) correlations among multi-constituents and multi-targets, integrating chemical profiling, ADME/PK processes, molecular network calculation and resulting experimental validation, the use of Integrative Pharmacology has become widespread. The data has provided a novel paradigm to evaluate the druggability of bioactive ingredients of herbs or formulae, to decipher the pharmacological mechanisms of drug action and to screen potentially new indications for approved drugs and previously unidentified adverse events. On this basis, Integrative Pharmacology may offer an effective way to test the potential scientific basis for traditional medicines and to assess what roles of traditional medicine can and cannot play in pharmaceuticals.
