Abstract: Epigenetic modifications of histone tails play an essential role in the regulation of eukaryotic transcription. Writer and eraser enzymes establish and maintain the epigenetic code by creating or removing posttranslational marks. Specific binding proteins, called readers, recognize the modifications and mediate epigenetic signalling. Here, we present a versatile assay platform for the investigation of the interaction between methyl lysine readers and their ligands. This can be utilized for the screening of small-molecule inhibitors of such protein-protein interactions and the detailed characterization of the inhibition. Our platform is constructed in a modular way consisting of ort...

Between Wisdom and Torah

Previous scholars have largely approached Wisdom and Torah in the Second Temple Period through a type of reception history, whereby the two concepts have been understood as signifiers of independent, earlier “biblical” streams of tradition that later came together in the Hellenistic and Roman eras, largely under the process of a so-called “torahization” of wisdom. Recent studies critiquing the nature of wisdom and wisdom literature as operative categories for understanding scribal cultures in early Judaism, as well as newer approaches to conceptualizing Torah and authorizing-compositional practices related to the Pentateuchal texts, however, have challenged the foundations on which t...

The multidisciplinary science of chemical proteomics studies how small molecules of synthetic or natural origin bind to proteins and modulate their function. In Chemical Proteomics: Methods and Protocols, expert researchers in the field provide key techniques to investigate chemical proteomics focusing on analytical strategies, how probes are generated, techniques for the discovery of small molecule targets and the probing of target function, and small molecule ligand and drug discovery. Written in the highly successful Methods in Molecular BiologyTM series format, chapters include introductions to their respective topics, lists of the necessary materials and reagents, step-by-step, readily reproducible laboratory protocols, and key tips on troubleshooting and avoiding known pitfalls. Authoritative and practical, Chemical Proteomics : Methods and Protocols seeks to provide methodologies that will contribute to a wider application of chemical proteomics methods in biochemical and cell biological laboratories.

Living I Was Your Plague

From the author of the acclaimed biography Martin Luther: Renegade and Prophet, new perspectives on how Luther and others crafted his larger-than-life image Martin Luther was a controversial figure during his lifetime, eliciting strong emotions in friends and enemies alike, and his outsized persona has left an indelible mark on the world today. Living I Was Your Plague explores how Luther carefully crafted his own image and how he has been portrayed in his own times and ours, painting a unique portrait of the man who set in motion a revolution that sundered Western Christendom. Renowned Luther biographer Lyndal Roper examines how the painter Lucas Cranach produced images that made the reform...
