Aquaporins

The aquaporin field has matured at an exceptionally fast pace and we are at the verge to develop serious strategies to therapeutically modulate aquaporin function directly or via regulatory networks. Key prerequisites are available today: i. a considerable (and growing) number of aquaporin crystal structures for the rational design of inhibitory molecules, ii. elaborate molecular dynamics simulation techniques for theoretical analyses of selectivity mechanisms and docking experiments, iii. comprehensive data on aquaporin immunohistochemistry, iv. aquaporin knockout animals for physiological studies, and v. assay systems for compound library screenings. The structure of this volume on aquaporins follows the points laid out above and thus covers the developments from basic research to potential pharmacological use. Situated between pharmacology textbooks and recent scientific papers this book provides a timely overview for readers from the fundamental as well as the applied disciplines.

Abstract: Background and Aims Autosomal dominant polycystic liver and kidney disease is a spectrum of hereditary diseases, which display disturbed function of primary cilia leading to cyst formation. In autosomal dominant polycystic kidney disease a genetic cause can be determined in almost all cases. However, in isolated polycystic liver disease (PLD) about half of all cases remain genetically unsolved, suggesting more, so far unidentified genes to be implicated in this disease. Methods Customized next-generation sequencing was used to identify the underlying pathogenesis in two related patients with PLD. A variant identified in SEC61A1 was further analysed in immortalized patients' urine s...

PATJ Inhibits Histone Deacetylase 7 to Control Tight Junction Formation and Cell Polarity

Abstract: The conserved multiple PDZ-domain containing protein PATJ stabilizes the Crumbs-Pals1 complex to regulate apical-basal polarity and tight junction formation in epithelial cells. However, the molecular mechanism of PATJ's function in these processes is still unclear. In this study, we demonstrate that knockout of PATJ in epithelial cells results in tight junction defects as well as in a disturbed apical-basal polarity and impaired lumen formation in three-dimensional cyst assays. Mechanistically, we found PATJ to associate with and inhibit histone deacetylase 7 (HDAC7). Inhibition or downregulation of HDAC7 restores polarity and lumen formation. Gene expression analysis of PATJ-defi...

Branching Morphogenesis

Branching morphogenesis, the creation of branched structures in the body, is a key feature of animal and plant development. This book brings together, for the first time, expert researchers working on a variety of branching systems to present a state-of-the-art view of the mechanisms that control branching morphogenesis. Systems considered range from single cells, to blood vessel and drainage duct systems to entire body plans, and approaches range from observation through experiment to detailed biophysical modelling. The result is an integrated overview of branching.

Principles of Tissue Engineering

Now in its fifth edition, Principles of Tissue Engineering has been the definite resource in the field of tissue engineering for more than a decade. The fifth edition provides an update on this rapidly progressing field, combining the prerequisites for a general understanding of tissue growth and development, the tools and theoretical information needed to design tissues and organs, as well as a presentation by the world's experts of what is currently known about each specific organ system. As in previous editions, this book creates a comprehensive work that strikes a balance among the diversity of subjects that are related to tissue engineering, including biology, chemistry, material scienc...

This book provides a comprehensive account of vascular biology and pathology and its significance for health and disease. It systematically and chronologically explains how we came to our current understanding of the vasculature and it ́s function today, and describes in an entertaining way the diverse flaws and turns in science and medicine from the past. It thereby offers a complete and well-studied history on vascular biology and medicine. The book has an easy-to-read style and is written for students as well as scientists, physicians and lecturers in the field of biomedicine, human physiology, cardiology and hematology.
